Pharmacokinetics of triple-combination antihypertensive drugs. Active substance profiles following oral administration and 24-hour blood pressure control Review article
Main Article Content
Abstract
Triple single-pill combinations (SPCs) bring together substances with markedly different pharmacokinetic profiles. This paper reviews three combinations used in arterial hypertension: perindopril arginine + indapamide + amlodipine, telmisartan + amlodipine + hydrochlorothiazide, and amlodipine + valsartan + hydrochlorothiazide. For each substance, Tmax, half-life, bioavailability, absorption and elimination, food effect and time to steady state are presented, together with modelled concentration-time curves after a single dose and during repeated dosing. Each combination shows the same kinetic complementarity: a short-Tmax component provides rapid onset, while a long half-life component sustains exposure to the end of the 24-hour dosing interval.
